De Fujifilm GFX 100S is een Medium Format systeemcamera met een 102MP 43,8 x 32,9 mm BSI CMOS-sensor. De camera is speciaal ontworpen voor professionals die beeldkwaliteit en het printen van werk belangrijk vinden. De camera schiet 16-bit raw foto's met een breed dynamisch bereik en gevoeligheidsbereik van ISO 50-102400.
Door gebruik te maken van de volledige breedte van de sensor, is DCI en UH 4K video-opname mogelijk op 4:2:0 10-bit intern of 4:2:2 10-bit via HDMI en ProRes Raw 12-bit in combinatie met een external recorder zoals de Atomos Ninja V. Verder heeft de camera ingebouwde beeldstabilisatie dat tot wel 6 stops kan corrigeren.
Ten slotte heeft de camera een LCD touchscreen, twee UHS-II SD kaartslots, weegt de camera slechts 900 gram en zijn de afmetingen 150 x 104.2 x 87.2 mm.
